A Critical Appraisal of the Threshold of Toxicity Model for Non-Carcinogens
نویسنده
چکیده
Most regulatory agencies assume that there is no safe level of exposure to carcinogens but that a threshold, or “safe” exposure level exists for non-carcinogens. However, recent discoveries have cast serious doubt on the validity of this concept. Five examples of non-carcinogens without an apparent threshold (neonicotinoids, dioxin, dieldrin, endocrine disruptors, and sulfhydryl-reactive metals) are presented. It is also clear by now that the threshold model for non-carcinogens may seriously underestimate actual risk. Risk assessments can no longer assume thresholds for non-carcinogens when the shape of the dose-response curve is linear at low concentrations. Risk management of such chemicals should be based on the ALARA principle (“as low as reasonably achievable”). *Corresponding author: Henk A Tennekes, Experimental Toxicology Services (ETS) Nederland BV, Frankensteeg 4, 7201KN Zutphen, The Netherlands, Tel: +31(0)575547717; E-mail: [email protected] Received September 22, 2016; Accepted September 28, 2016; Published September 30, 2016 Citation: Tennekes HA (2016) A Critical Appraisal of the Threshold of Toxicity Model for Non-Carcinogens. Some Notes on Critical Appraisal of Prevalence Studies; Comment on: “The Development of a Critical Appraisal Tool for Use in Systematic Reviews Addressing Questions of Prevalence”
Decisions in healthcare should be based on information obtained according to the principles of Evidence-Based Medicine (EBM). An increasing number of systematic reviews are published which summarize the results of prevalence studies. Interpretation of the results of these reviews should be accompanied by an appraisal of the methodological quality of the included data and studies. The critical a...
متن کاملEffect of Cell Size and Shape on Electric Field Threshold and Critical Transmembrane Voltage for Electroporation
Introduction: Electroporation is a technique for increasing the permeability of the cell membrane to otherwise non-permeate molecules due to an external electric field. This permeability enhancement is detectable if the induced transmembrane voltage becomes greater than a critical value which depends on the pulse strength threshold. In this study, the variabil...

متن کاملThe Development of a Critical Appraisal Tool for Use in Systematic Reviews: Addressing Questions of Prevalence
Background Recently there has been a significant increase in the number of systematic reviews addressing questions of prevalence. Key features of a systematic review include the creation of an a priori protocol, clear inclusion criteria, a structured and systematic search process, critical appraisal of studies, and a formal process of data extraction followed by methods to synthesize, or combin...
